Culture

Embryos are grown in single step media with oil overlay to maintain a stable culture system and preserve embryo viability, which is key to improve clinical outcomes.

Oocytes submitted to ICSI, are transferred to the culture dish the same day after injection. On the other hand, oocytes submitted to conventional IVF are checked for fertilization outcome on day 1 and transferred from the Fertilization media dish to the culture dish per se.

Culture media needs to support embryo development through early (day 3) and blastocyst (day 5/6) stages. Extended culture can even be performed until day 7 of development. Embryos that reach blastocyst stage with better embryo quality will have better chances of implantation.

For this purpose, embryos are grown in Single Step Culture Media with Hypure® Oil overlay. Hypure® Heavy Oil is a good option for extended culture with no dish replacement on day 3.

The composition of culture media is important when trying to improve current culture conditions

Kitazato Single Step Culture Media

Offer a perfect balance of organic and inorganic salts, energy substrates and a great assortment of amino acids. Amino acids support numerous cellular processes, including acting as metabolites, osmolytes, antioxidants, and buffers, and likely help alleviate stress in the embryo culture system.

Kitazato Single Step Culture Media re supplemented with HSA, this macromolecule can act as:

  • Surfactant
  • Nitrogen Source
  • Stabilize membranes and modulate the physical microenvironment
  • Act as a carrier molecule for other compounds, or even bind trace element and toxins

It is clear that maintenance of a stable culture environment is advantageous when attempting to minimize environmental perturbations within the culture system that may negatively impact the developing embryo.

The composition of the medium used to grow preimplantation embryos in vitro is certainly important in preserving embryo viability and Kitazato Single Step Culture Media help ensure consistency among laboratories.

Kitazato Hypure® Heavy Oil used as an overlay, allows small volumes of medium to be used as it prevents the evaporation of the media, whilst maintains stable osmolality and pH in the culture medium. Optimizing the embryo culture system, promises to improve the overall success of IVF, allowing routine elective single ET and thereby yielding ideal perinatal outcomes (1).
